so I got me some pre sterilized agar plates on the net. But I'm going to sterilize it again just to be sure. At what PSI and for how long would I keep it there for? Tia
Use this link, section on sterilization of culture media, to determine your sterilization temp and duration. Since yours are presterilized, I wouldn't go over 20 min. At 121oC, or the equivalent recommended temp/duration pair if you want to try higher temps for shorter periods.
Then you can convert the temp to Fahrenheit, and use this link to determine your operating pressure based on your required temperature. The resulting "steam pressure" is the pressure you want to shoot for.
Remember that proper purging of your pressure cooker is essential in order for these calculations to work. Air and other non-condensable gasses can really fuck up your pressure-temperature relationship.
