Monoamine Oxidases (MAOs) are enzymes that are typically bound to mitochondrial surfaces, and serve the function of deactivation (through oxidation) of various  amines that act as neurotransmitters. While it is true that MAOs are involved in the regulation of amines taken up by digestion, it does not act in the GI tract to prevent the various digestive mechanisms. MAOs act on the existing amines that are already absorbed from the GI contents. MAOs are found in high amounts in the brain and heart.

Monoamine Oxidase Inhibitors (MAOIs) inhibit this action, as the name implies. This can be used to potentiate the effects of certain drugs that are quickly consumed by MAOs, which is why natural sources of MAOIs are used in oral DMT formulations, where the uptake rate of DMT is slow and normally outpaced by the action of the MAOs.

However, MAOIs also disrupt the regulation of amines taken up through the GI tract, as well as regulation of neurotransmitters in the brain. This can present significant risk if not carefully planned out to avoid certain foods or compounds.

The main receptors targeted by the compounds in Kratom are the opioid receptors, where it performs some varying degree of agonism (activation). The strongest action is focused on the kappa-opioid or k-opioid receptor. Studies have shown potential for k-opioid agonism as a treatment for addiction, which combined with the partial agonism of the mu and delta receptors, explains how kratom addiction is harder to achieve, and why kratom withdrawal is usually not as severe as standard opioid addiction and withdrawal.

However, the long-term (>30 min) addiction treatment potential of k-receptor agonism is dependent on the accompanying signal transduction and neurotransmitter regulation actions triggered by the k-receptor agonism. I suspect that by pairing Kratom with Harmala extract, you are bypassing the anti-addiction benefits of Kratom's affinity for k-opioid agonism, while simultaneously decreasing the body's ability to regulate the amines and their associated neurotransmitter releases (e.g. dopamine in the reward pathway), and increasing the longterm effects on the brain that result in addiction and withdrawal.

I think I've read somewhere that some peripheral antagonism between Kratom compounds and the 5-HT2a receptor may result in blood pressure increases, and that these effects can be amplified by combining a MAOI with Kratom. You should probably approach this combo with caution...

Lol I literally typed up what I had laying around from previous research, I take my substances and their effects on my body seriously, and do plenty of research prior to dosing up on anything. MAOIs are of particular interest since they can kill you just by eating the wrong food while on them. Opiates and those substances that act on opioid receptors are also of particular interest, as I have experienced how quickly a couple of weeks of fentanyl and oxycodone administration in the hospital can lead to pretty awful withdrawal symptoms.

Then I read up on what's going on, and anything that interests me tends to stick in my head. I have lots of information stored there, but none of it is particularly useful...:shrug:

That's why I come here to bother you with it :dancingshroom: that and to administer the entertaining rants under PBs account :nyan:

I got IV fentanyl in the hospital and while enjoyable I got the impression that to keep a habit going for any amount of time you would eventually have to redose about every two hours and even then the high would not be that good.  Seems like you would just end up chasing your tail.

I have :tinfoil: theories on opiates..:awesketch:

ive never tried fentanyl but I would think dilaudid is way better for chronic pain

I was on a fentanyl drip for about 3 weeks while I was in traction for a major fall injury a decade or two ago, then on liquid oxy for several weeks after I came home.

The whole experience turned me off of opiates...I hate that feeling of needing something that I know I don't.

Unless its caffeine, then I 100% can't live without it, and will die if deprived of it for more than 24 hours...

yeah man when I had dilaudid for a kidney stone I was ain free and happy in reno qiyh the sun coming up, I had insomnia at night nx dad took me to ER, they gave me hydros which I fundoturnatly abused and now he owont even give me Xanax because of past abuse and dependence...……...:mushroom2:

I’ve always kind of figured, considering it’s used for what it’s used for there may be some incentive to deliberately not develop a tolerance or familiarity to opiates.  Granted I t&w kratom all day but I feel pretty certain an oxycontin would still knock me on my ass if I were to take a solid dose.

Did you enjoy the high at all or dislike it and want to get it out of your system asap?

Some make me feel good and some I get sweaty and ill.  I would bet the ones that make me ill would start working once I took them for awhile and got a tolerance going.  Vicodin = wonderful, Percocet = hurl.

I'm not a good judge on opiates. I'm allergic to morphine (or rather, morphine displaces histamine from their storage areas in the cells for me) so I itch like a motherfucker when I'm on any opiate, and morphine will make me want to tear my skin off.

Aside from that, I'm not a fan of the CNS depression, the warm blanket feeling isn't worth the withdrawal IMO :shrug:
