I think you're over-thinking it. It looks pretty healthy to me. Those tiny first leaves are the cotyledons. They are the embryonic first leaves of a seedling and are commonly shed by certain strains after the second to fourth set of true leaves have formed.

If anything she may be a little hungry. Wait until the soil dries out pretty good, then give her a light feeding.

I do see what you're talking about. The yellowing on the little one-bladed leaf right above the cotyledon. I wouldn't count that as a true leaf either. You see a lot of seedlings that are started with the light too high and they don't get the single blade leaf. It looks like you kept your light at the right height. The yellowing is what made me suggest a small feeding, but wait until it dries out again first. It looks good.

looks like you could need cal-mag. your plant looks like its not absorbing nutrients properlly. RO water has zero calcium, mag., it has virtually nothing in it. FYI RO water can be more difficult to deal with, I have found that when growing in dirt its best to use tap water or rain water both have trace elements in it. I would give it a low dose of cal-mag with tap water and a week to recover and you should be fine.
